NF-κB: The Master Inflammatory Switch
Nuclear factor kappa B (NF-κB) is a transcription factor that acts as the master regulator of inflammatory gene expression. When chronically activated — by stress, poor diet, environmental toxins, or aging — it drives the production of pro-inflammatory cytokines including TNF-α, IL-1β, and IL-6. Curcumin, the active compound in turmeric, is one of the most potent natural inhibitors of NF-κB identified to date.
Omega-3s and the Resolution of Inflammation
Inflammation resolution is an active biological process, not merely the absence of inflammation. EPA and DHA — the long-chain omega-3 fatty acids found in marine sources — are precursors to specialized pro-resolving mediators (SPMs) including resolvins and protectins. These molecules actively signal the immune system to stand down. Most people are severely deficient in EPA and DHA, which impairs this resolution pathway.
Oxidative Stress and the Inflammatory Cycle
Reactive oxygen species (ROS) and chronic inflammation exist in a self-reinforcing cycle: oxidative stress activates inflammatory pathways, and inflammation generates more oxidative stress. Polyphenols — including EGCG from green tea, resveratrol, and quercetin — interrupt this cycle by neutralizing free radicals and activating the Nrf2 antioxidant response pathway, which upregulates the body's own antioxidant defenses.
